● Methods for obtaining periapical biopsies in cases that fail after root canal treatment.
● Methods for obtaining tooth biopsies. Preparation of tooth to expedite penetration of the fixative in the pulp.
● Fixation of tissue. Types of fixatives.
● Protocols for demineralization.
● Infiltration of specimens in paraffin. Preparation of the paraffin block.
● Microtomy. Serial sectioning procedures.
● Staining with Hematoxylin & Eosin. Techniques to stain bacteria (Taylor modification of the Brown & Brenn technique).
● Observation of the stained sections in the light microscope. Recognition of cells and structures of the normal pulp. Recognition of cells and structures of periapical tissues. Recognition of inflammatory cells and regressive changes. Recognition of bacteria.
● Taking photomicrographs.
